Description
The Bradford assay is a simple and rapid method for measuring total protein concentration based on the binding of Coomassie Brilliant Blue (CBB-G250) to proteins. Upon binding, the dye shifts in color from brown (465 nm) to blue (595 nm), and protein concentration is determined by measuring absorbance at 595 nm using a standard curve.
However, the Bradford assay is sensitive to surfactants, which can interfere with dye–protein binding and affect accuracy. The WSE-7510 EzBradford Protein Assay Kit minimizes surfactant interference and enables reliable protein quantification in the ranges of 10–2,000 μg/mL (standard assay) and 1–50 μg/mL (low-range assay).
The results show protein quantification using the EzBradford Protein Assay Kit with BGG standard dilution series supplemented with either 0.5% SDS (BGG_SDS) or 20 mM DTT (BGG_DTT). No inhibitory effect was observed with DTT. However, BGG containing 0.5% SDS affected color development, resulting in inhibited quantification. When Protein Diluent was added to SDS-containing BGG (BGG_SDS_Diluent), the inhibitory effect of SDS was reduced, and protein quantification showed good linearity comparable to BGG without additives.
These results demonstrate that the EzBradford Protein Assay Kit enables reliable protein quantification by minimizing interference from surfactants.
To evaluate the stability of color development using the EzBradford Protein Assay Kit, absorbance was measured every 2 minutes for 1 hour. The results showed that, regardless of protein concentration, the measured values remained constant, indicating that the color reaction is stable for approximately one hour after the reaction begins.
The table below summarizes the permissible concentrations of coexisting substances in sample solutions that do not affect the reaction system of protein quantification reagents. Please note that, depending on the type and concentration of protein, the combination of reagents, and conditions such as pH, the reaction system may still be affected even at concentrations lower than those listed in the table.
