Purpose and Application

  • “cp-PAGEL Neo” is a high-performance, ready-to-use polyacrylamide gel (glass plate size: 60 mm × 60 mm) for the separation of low molecular weight proteins.

Experimental Conditions

  • Gel: cp-PAGEL Neo (CPN-16.5S)

  • Electrophoresis buffer: 1× EzRunT (Tris-Tricine-SDS)

  • Electrophoresis system: Compact PAGE Neo

  • Staining method: EzStain AQua (rapid staining protocol)

Post-electrophoresis staining

  • After separation with cp-PAGEL Neo, the gel was stained either with EzStain AQua (a rapid staining method) or with CBB staining solution (0.25% CBB, 40% methanol, 10% acetic acid) following fixation.
  • Bands at 1.0, 2.5, and 3.5 kDa—highlighted by the dotted area—were detected only with EzStain AQua.
  • To effectively visualize low molecular weight proteins, it is essential to perform staining and destaining as quickly as possible.
  • Since EzStain AQua is alcohol-free, it minimizes gel deformation due to dehydration. Additionally, its short staining time reduces protein diffusion, making it highly suitable for detecting low molecular weight proteins.

* Gel staining method

1. Standard Method

  • Immerse the gel in EzStain AQua and gently shake for at least 3 hours.

  • Replace the solution with distilled water and destain until the background becomes transparent.

2. Rapid Method

  • Immerse the gel in EzStain AQua, then heat in a microwave (600 W) for 1 minute.

  • Gently shake for 10 to 60 minutes to allow staining.

  • Replace with distilled water, heat in the microwave again, and then shake at room temperature for 30 to 60 minutes to destain until the background becomes transparent.

* For low molecular weight protein separation, use Tris-Tricine-SDS buffer (AE-1415 EzRunT).
* cp-PAGEL Neo is a ready-to-use gel designed for Tricine-PAGE. For SDS-PAGE or Native PAGE, use c-PAGEL Neo in combination with AE-1410 EzRun (SDS-PAGE buffer) or WSE-7055 EzRunTG (SDS-free buffer), respectively.
* We recommend storing the gel at a temperature between 5°C and 10°C. If it is placed near the cold air outlet of the refrigerator, the temperature may drop below 4°C, which can cause the gel to freeze and become unusable.
